培(pei)養基介紹(shao),P2RX7基因的(de)遺(yi)傳(chuan)性變(bian)異(yi)會(hui)影響到(dao)不(bu)同人(ren)對疼(teng)痛(tong)的感受。因為(wei)疼(teng)痛(tong)感知的(de)個(ge)體差(cha)異(yi)性和(he)對(dui)疼(teng)痛(tong)緩(huan)解(jie)藥(yao)物(wu)的(de)反(fan)應不(bu)同,疼(teng)痛(tong)相關臨(lin)床試(shi)驗的(de)進(jin)行(xing)壹直(zhi)都存在壹定難(nan)度(du),這(zhe)項研(yan)究(jiu)或(huo)能有助我們在該(gai)基因變(bian)異的基礎上(shang)開發出(chu)相應的個(ge)體疼(teng)痛(tong)療(liao)法(fa)。
培(pei)養基指出(chu),Jeff Mogil、Michael Salter等人(ren)觀察了(le)大(da)量小鼠的(de)不(bu)同疼(teng)痛(tong)敏(min)感度(du),註意(yi)到小鼠P2RX7基因的(de)突變與這(zhe)些(xie)差(cha)異(yi)有關。他(ta)們發現該(gai)基因的(de)某(mou)壹變(bian)異(yi)讓(rang)壹種(zhong)具(ju)有細(xi)胞(bao)膜中(zhong)離(li)子通道作(zuo)用(yong)的受(shou)體P2X7生成大(da)孔(kong)徑(jing);產生該(gai)基因變(bian)異的小鼠比(bi)未(wei)產生變(bian)異的(de)小鼠會(hui)表現(xian)出(chu)更(geng)多疼(teng)痛(tong),而壹種(zhong)能阻礙該(gai)大(da)孔(kong)徑(jing)生(sheng)成(cheng)的(de)縮氨(an)酸(suan)則(ze)可(ke)以減(jian)輕變異(yi)小鼠的(de)疼(teng)痛(tong)。之後(hou),Salter和同事將該(gai)發(fa)現應用(yong)於人(ren)體試(shi)驗,他(ta)們發現如(ru)果P2RX7基因的(de)變異不(bu)導(dao)致(zhi)P2X7受(shou)體生成大(da)孔(kong)徑(jing)的(de)話(hua),那(na)麽(me)乳(ru)房(fang)切(qie)除術後(hou)疼(teng)痛(tong)和關節(jie)炎(yan)疼(teng)痛(tong)的患(huan)者(zhe)會(hui)表現出(chu)較少的(de)疼(teng)痛(tong)。
